Protocol For The Quantitative Analysis Of Adulteration Of Basmati Rice With Sherbati, Mugad Sugandha, Pak 386 Or Superfine

Scope

Quantitative detection of Basmati 370, Dehradun (Type 3), Basmati 217, Taraori, Ranbir Basmati, Kernel, Basmati 386, Pusa Basmati and Super Basmati. The microsatellite marker RM201 has been chosen to quantify the extent of adulteration of these premium Basmati varieties with Sherbati, Mugad Sugandha, Pak 386 or Superfine.

The test is based on the ratio of alleles of RM201 present in the sample, determined by measuring the areas under the peaks in a capillary sequencer for all peaks associated with each allele (including the main peak, stutter peaks and + A peaks).

The test is quantitative, i.e. it can detect whether a consignment contains a pure variety or a certain range of mixtures with some known adulterants. It is not applicable to all present and possible adulterant varieties, for which different microsatellite markers, or other types of markers, may be more appropriate. A separate operating procedure is available for variety identification.
